Generating C-terminal fusions with Gateway

 

 

 

 

 

The entry clones are then used in a "two-way multisite" LR reaction. In this case we use a different Destination vector than the one used for generating N-terminal fusions. Here, the Dest vector has an attR1 and attR3 site. This forces the recombination to occur as described above - resulting in the proper orientation of the 5' and 3' entry clone into the pCS-based Dest vector to yield an ORF-egfp injection construct.
