Gateway cloning overview


Gateway cloning is a recombination based method of generating plasmid constructs (Hartley et al. 2000; Walhout et al. 2000). Gateway cloning entails two different recombination steps: 1. BP recombination - this is used to generate an "ENTRY" clone. 2. LR recombination - transfer of your ORF from an ENTRY clone into any of a number of "Destination" vectors for functional analyses. Although this is an incredibly useful approach for generating constructs, the application of this technology to zebrafish requires the availbility of appropriate Destination vectors. We have constructed a series of pCS- and Tol2-based plasmids that are compatible for Gateway cloning and can be used in zebrafish.
